Forkhead box protein O3 (FOXO3a), or FKHRL1, is a ubiquitous transcription factor that triggers the expression of pro-apoptotic genes under conditions of oxidative stress. FOXO3a is regulated by the Akt kinase which phosphorylates FOXO3a on serine 253, 315, and on threonine 32 residues. Modified FOXO3a is recognized by the 14-3-3 proteins which export FOXO3a to the cytoplasm, and consequently terminates its transcriptional activity. FOXO3a may also be regulated by lysine methylation in neuronal cells.
