Matrix metalloproteinase-24 (UniProt: Q9R0S2; also known as MMP-24, Matrix metalloproteinase-21, MMP-21, Membrane-type matrix metalloproteinase 5, MT-MMP 5, MTMMP5, Membrane-type-5 matrix metalloproteinase, MT5-MMP, MT5MMP) is encoded by the Mmp24 (also known as Mmp21, Mt5mmp) gene (Gene ID: 17391) in murine species. MMP-24 is a single-pass type I membrane protein that is mainly expressed in neuronal cells of both central and peripheral nervous systems. It mediates the cleavage of N-cadherin and acts as a regulator of neuro-immune interactions and neural stem cell quiescence. It is also involved in cell-cell interactions between nociceptive neurites and mast cells. MMP-24 is synthesized with a signal peptide (aa 1-41) and a propeptide (aa 42-128) that are subsequently cleaved off to generate functional MMP-24 (aa 129-618) or processed MMP-24 (aa 129-554). MMP-24 contains a cysteine switch motif (aa 110-117) that binds the catalytic zinc ion and inhibits its enzyme activity. The dissociation of the cysteine from the zinc ion upon the activation-peptide release results in the activation of enzyme.
