Zinc finger homeobox protein 3 (UniProt: Q15911; also known as AT motif-binding factor 1, AT-binding transcription factor 1, ATBF1, Alpha-fetoprotein enhancer-binding protein, Zinc finger homeodomain protein 3, ZFH-3) is encoded by the ZFHX3 (also known as ATBF1) gene (Gene ID: 463) in human. ATBF1 contains 23 C2H2-type zinc fingers and 4 homeobox DNA-binding domains. It is shown to translocate from the cytoplasm to the nucleus following TGF-beta stimulation. It acts as a transcriptional regulator that can act as an activator or a repressor. It inhibits the enhancer element of the AFP gene by binding to its AT-rich core sequence. In concert with SMAD-dependent TGF-beta signaling, it can repress the transcription of AFP via its interaction with SMAD2/3. ATBF1 is shown to regulate myoblasts differentiation through the binding to the AT-rich sequence of MYF6 promoter and promoter repression. It is not found in normal gastric mucosa, but is found in gastric carcinoma cells and its expression is higher in ER-positive breast tumors than ER-negative breast tumors.
