The protein CUL4B acts as the scaffold protein within the multi-protein RING E3 ubiquitin protein ligase complexes needed for ubiquitination and subsequent proteasomal degradation of target proteins. CUL4B appears to contribute to the catalysis of ubiquitination by positioning the substrate target protein and the E3 ubiquitin ligase. Among the targets CUL4B is associated with include histone H2A, H3, p53, cyclin E and H4. The cell signaling mTORC1 pathway is also regulated via CUL4b interaction with MLST8 a key player in the mTORC1 pathway. Thus CUL4B is important is proper cell division, and cell cycle function, and is also associated with tumorigenesis in cancer by coordinating with various methylases in promoting specific DNA methylation-based epigenetic silencing in certain cancers. CUL4B is localized in the nucleus as well as the cytoplasm depending upon the context and is widely expressed.
