Neuron navigator 1 (UniProt Q8NEY1; also known as Pore membrane and/or filament-interacting-like protein 3, Steerin-1, Unc-53 homolog 1, unc53H1) is encoded by the NAV1 (also known as KIAA1151, KIAA1213, POMFIL3, STEERIN1, UNC53H1) gene (Gene ID 89796) in human. NAV1 is a member of the family of proteins called navigators that play a role in mediating neurite outgrowth. NAV1 is shown to interact with the guanine nucleotide exchange factor (GEF) TRIO, whereby it enhances the affinity of TRIO for Rho family GTPases Rac1 and RhoG. The plus-end-tracking protein (+TIP) EB1 at the ends of growing microtubules recruits TRIO, facilitating the formation and localization of functional NAV1-TRIO complexes, which in turn regulates neurite outgrowth by selectively activating Rac1 at the growth cone.
