TMSB15A protein is a key regulator of the cytoskeleton organization and polymerization, and it thus plays a role in cell migration, proliferation and cancer. TMSB15A does this because TMSB15A binds to and sequesters actin monomers and limits action polymerization. TMSB15A is differentially expressed in many tissues and cancer cell lines. Because of differential expression in certain cancers, for instance in triple negative breast cancer but not colon cancer, TMSB15A is being examined as a histochemical marker for measuring the effectiveness of cancer therapy on cancer. TMSB15A is also important in the nervous system and brain diseases. Notably, TMSB15A expression is increased upon brain stress and surprisingly TMSB15A localization differs depending upon the type of stress used and neuronal cell type. Because of its interaction with actin, TMSB15A is localized to the cytoplasm most of time, however, in the brain and when cells are placed under various stresses, TMSB15A can be translocated to the brain to perform yet unknown functions as well.
