Phosphofurin acidic cluster sorting protein 2 (PACS2), also known as PACS-2, PACS1-like protein, and encoded by the gene name PACS2/ KIAA0602/ PACS1L, is a amember of the PACS family. PACS2 is a multifunctional sorting protein that controls the endoplasmic reticulum (ER)-mitochondria communication, including the apposition of mitochondria with the ER and ER homeostasis. Additionally, PACS2 translocates BIB to mitochondria in response to apoptic inducers, which initiates a sequence of events including the formation of mitochondrial truncated BID, the release of cytochrome C, and the activation of caspase-3 thereby causing cell death. Recent studies have indicated that PACS2 may also be involved in ion channel trafficking, directing acidic cluster-containing ion channels to distinct subcellular compartments. PACS2 interacts with BID, PKD2, and HIV-1 Nef.
