The anterior foregut endoderm (AFE) gives rise to therapeutically relevant cell types in tissues such as the lung, esophagus, salivary glands, thymus, parathyroid and thyroid. Our cryopreserved anterior foregut endoderm (AFE) progenitor cells were derived from integration-free human iPS cells (Cat. No. SCC271) reprogrammed using the Simplicon RNA reprogramming technology (Cat. No. SCR550) in chemically defined/serum-free culture conditions. The AFE cells are double positive for Sox2 (A,C; Sigma AB5603A4; 1:50), Pax9 (B, C; Abcam ab28538; 1:200) and approximately 20-30% are double positive for EpCAM (D, F; Sigma MAB4444; 1:500) and Pax9 (E, F). The human iPSC derived AFE progenitors are a component of the 3dGRO(TM) Human Lung Organoid Culture System which can be used to differentiate human iPS cells into mature lung organoids that structurally resemble the in vivo branching airway and early alveolar structures.
