The AlphaLISA SureFire Ultra Human and Mouse Total Aggregate TDP-43 assay is a sandwich immunoassay for quantitative detection of total aggregate TDP-43 in cellular lysates using Alpha Technology. TAR DNA binding protein 43 (TDP-43) is a DNA and RNA-binding protein which plays a crucial role in RNA metabolism. Mainly located in the nucleus, TDP-43 shuttles between the nucleus and the cytoplasm. In pathological conditions, including stress or mutation, TDP-43 is mislocalized in the cytoplasm, which can be cleaved in C terminal fragments, thus leading to an accumulation of ubiquitinated and hyperphosphorylated toxic aggregates and then insoluble inclusion bodies. Aggregated TDP-43 has been identified as a hallmark of amyotrophic lateral sclerosis (ALS) and frontotemporal lobar dementia (FTLD), and more widely in several neurodegenerative diseases such as Alzheimer's disease.
