Ghrelin exists in two main forms, an acylated (active) and deacylated (inactive) form. Ghrelin is derived from a pre-pro form after several stages of enzymatic processing and is secreted when the stomach is empty. Ghrelin acts to increase hunger (appetite) and to ensure stomach acid preparedness for food intake. Ghrelin has been shown to play a role as a reward molecule by interacting with dopaminergic neurons in the brain. Interestingly, leptin seems to have a contradictory role to ghrelin where it suppresses appetite and reward pathways. Ghrelin is being intently studied for its role in the regulation of appetite, sleep and circadian rhythm, its anti-inflammatory effect on the gastrointestinal tract, and depression. This AlphaLISA detection kit allows for the detection of total ghrelin levels in serum, plasma, and cell culture supernatants. AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
