One antibody of interest is labeled with a donor fluorophore (a LANCE Europium chelate) and the second molecule is labeled with an acceptor fluorophore (ULight™ dye). Upon excitation at 320 or 340 nm, energy can be transferred from the donor Europium chelate to the acceptor fluorophore if sufficiently close for FRET (~10 nm). This results in the emission of light at 665 nm. Adiponectin (Acrp30, in mouse) is a hormone secreted by differentiated adipocytes that regulates energy homeostasis and glucose and lipid metabolism. It is expressed as a ~35 kDa protein. It has been demonstrated that Adiponectin / Acrp30 plays a major role in controlling whole-body metabolism, particularly by enhancing insulin sensitivity in muscle and liver, and by augmenting the oxydation of fatty acids in muscle.
